Schematic representation of the experimental procedure for identifying host factors involved in filovirus genome replication and transcription
A genome-wide siRNA screen was performed using 64,755 siRNAs and a mini-genome optimized for high-throughput screening. Results were analyzed by bioinformatics, and, based on these results, a further 696 siRNAs were tested in the same assay. The 79 best siRNAs were subsequently tested for activity during infection with a recombinant Ebola virus expressing luciferase. 23 host factors have been identified that appear to play a role in genome replication and transcription. © 2018, Thomas Hoenen, FLI.
